Docetaxel chemotherapy was administered to patients with metastatic castration-resistant prostate cancer (CRPC) for a decade, and demonstrated ability to improve the median overall survival by around 3 months 5,6. Currently, there is no effective therapy for recurrent prostate cancer; hence, a novel therapeutic method for CRPC is needed. By activating AMPK,AICAR promotes the switch from glycolysis to fatty acid oxidation, providing a more efficient energy source during prolonged exercise. Studies have demonstrated that AICAR can mimic the effects of exercise by enhancing muscle endurance and performance.

AMPK-dependent and independent effects of AICAR and compound C on T-cell responses

This leads to the alteration of the normal splicing and the inefficient production of full-length transcripts. Thus, about 85–90 % of SMN2 transcripts lack exon 7 and produce a truncated and unstable protein (SMNΔ7), which is rapidly degraded and therefore not functional 6. The remaining (~10–15 %) transcripts derived from the SMN2 produce a FL-SMN protein, which is only able to compensate partially for the absence of SMN1. SMN2 copy number is inversely correlated with the severity of the disease 7, 8, and usually the most affected patients are those with fewer copies of SMN2. Bile duct ligation (BDL) is an experimental animal model for extrahepatic bile duct obstruction that entails the ramification of small bile ducts within liver tissue accompanied by leukocytes infiltration and subsequent fibrosis 4.

Our data clearly support the hypothesis that the full capacity of AICAR to reduce insulin resistance requires its inflammation-suppressing ability as an essential component, in addition to other beneficial effects including lipid and glucose metabolism. Although AICAR/Compound C have been commonly used as an agonist/antagonist of AMPK, respectively, whether or not they are able to activate/inhibit AMPK in T cells remains unclear 18, 30, 34. Our previous data demonstrated that AMPK is specifically deleted in T cells from CD4-Cre+ AMPKα1fl/fl (KO) mice, but is intact in T cells from CD4-Cre- AMPKα1fl/fl (WT) mice 10. We thus continued to use this model to dissect the effects of AICAR/Compound C on AMPK in T cells. We first measured the AMPK activation using resting T cells from lymph nodes of WT and KO mice.

5.7. Population analysis

Spinal muscular atrophy (SMA) is a genetic neuromuscular disorder characterized by spinal and brainstem motor neuron (MN) loss and skeletal muscle paralysis. Currently, there is no effective treatment other than supportive care to ameliorate the quality of life of patients with SMA. Some studies have reported that physical exercise, by improving muscle strength and motor function, is potentially beneficial in SMA. The adenosine monophosphate-activated protein kinase agonist 5-aminoimidazole-4-carboxamide-1-β-D-ribofuranoside (AICAR) has been reported to be an exercise mimetic agent that is able to regulate muscle metabolism and increase endurance both at rest and during exercise.

AMPK and Akt Phosphorylate TBC1D1—Insulin stimulation activates Akt, AICAR stimulation activates AMPK, and muscle contraction activates both AMPK and Akt. To test this hypothesis, TBC1D1 was immunoprecipitated and incubated in vitro with recombinant AMPK, Akt, or AMPK plus Akt for 30 or 60 min. Incubation with AMPK, Akt, or AMPK and Akt combined resulted in similar increases in TBC1D1 PAS phosphorylation (Fig. 7B). No differences were observed between 30- and 60-min incubations, indicating maximal TBC1D1 PAS phosphorylation was achieved by 30 min. TBC1D1 phosphorylation by AMPK, but not Akt, caused a distinct upward shift in electrophoretic mobility similar to AICAR and contraction stimulation, which activate AMPK in vivo (Fig. 7A). Together, these findings suggest that AMPK may phosphorylate TBC1D1 at multiple sites in vivo and in vitro.

Image and Morphometric Analysis

In soleus muscle, immunodepletion of AS160, but not TBC1D1, resulted in nearly complete PAS-160 depletion. Accordingly, the time courses of PAS-160 phosphorylation in soleus (Figs. 2A and 3A) and tibialis anterior (Figs. 2B and 3B) muscle may represent the time courses of AS160 and TBC1D1 PAS phosphorylation, respectively. Insulin stimulates AS160 phosphorylation and increases glucose uptake by removing the brake effect of AS160 on GLUT4 exocytosis (8).

Among these mice, one group fed a Dox diet for 8 weeks was followed by a regular diet for 2 weeks (EG8OFF2). Many genetic studies support the notion that macrophage inflammation is a key component of obesity-induced inflammation and insulin resistance 5, 6, 7, 8. However, the fundamental mechanisms responsible for the altered inflammatory programs in obesity remain elusive. AICAR, short for 5-aminoimidazole-4-carboxamide ribonucleoside, is a short peptide that plays a role in energy homeostasis and a number of metabolic pathways. AICAR plays a role in the regulation of insulin receptors and how muscle cells function with regards to insulin. AICAR is under active investigation for its cancer-fighting properties and for its ability to protect heartIcardiovascular tissue. Baseline glucose values were measured in the animals on empty stomachs before insulin administration, as well as in dynamics 20, 40, 60, and 120 min after subcutaneous administration of insulin at a dose of 2 IU/kg.

The lysates were then centrifuged at 20,000 g for 10 min at 4 °C and an equal amount of 2× SDS sample buffer was into each supernatant. To analyze the levels of p-AMPKα and p-ACC in liver, the freshly excised liver tissues were added with ice cold lysis buffer (10 μl/mg liver weight), followed by homogenization and centrifugation as described above. The lysates were then mixed with 2× SDS sample buffer and then subjected to immunoblotting. Levels of total proteins and the levels of phosphorylation of proteins were analyzed on separate gels and representative images were shown. The band intensities on developed films were quantified using Image J software (National Institutes of Health Freeware). We postulated that upon starvation for glucose only, basal AMP might act as a necessary cofactor for AMPK activation via the lysosomal pathway, whereas elevated AMP may bind to additional sites on AMPK.

Point mutations on AMPK-γ1 (D317A), AMPK-γ2 (R531G), and AMPK-β1 (G2A) were made using a PCR-based site-directed mutagenesis method using PrimeSTAR HS polymerase (Takara). Expression plasmids for various proteins were constructed in the pcDNA3.3 vector for transient transfection (ectopic expression), in pBOBI for lentivirus packaging (stable expression). The lentivirus-based vector pLL3.7 was used for expression of siRNA in HEK293T and HEK293 cells. Hearts were cut into 1 mm3 pieces and digested with Liberase TH Roche Diagnostics, Indianapolis, IN). The resulting non-myocyte cells were immediately used for flow cytometry analysis 6 or for tissue culture.
